The reality star, 28, is said to be the producers' favourite, reports TMZ, thanks to a top notch screen test.
A "well-connected" source told the gossip site: "There's at least an 85 per cent chance she'll get the job."
Show bosses are said to be keen to exploit the huge success of Keeping Up With The Kardashians by picking Khloe for the role.
The aim is to appeal to a younger audience and the youngest Kardashian would join judges Demi Lovato and Britney Spears in a hipper line-up.

Kooky Khloe is married to NBA basketball player Lamar Odom and we reported back in June that the pair are undergoing fertility treatment.
Khloe allegedly visited a clinic in Beverly Hills to discuss her options after big sis Kourtney announced she was expecting her second child.
A source said: "She's thrilled for Kourtney, but she's really sensitive. And it's hard to see your sister get something you want so badly."
The star has previously admitted she feels under pressure to have a baby.
Basketball player Lamar already has two children from a previous relationship — Destiny, 13, and Lamar Jr, 10.
